Thanks for response. Unetbootin did not worked for me. PC has no floppy support. So I could not put MBR on the floppy. At first I did not want to modify MBR of PC. @Monsie Please note that at first when I installed puppy on USB using YAMI from pendrivelinux , it booted. After loading initrd pc reboots. So bios is capable to boot from usb. Try checking to see if the flashdrive has a bootflag set in gparted If not, set it and try again I checked with gparted, boot flag set. PLOP should work IF the flashdrive is bootable Test it in a different PC, with USB bootability to be sure... I tried this usb with plop, on a different pc, laptop ...

Hi, @bigpup @playdayz Thank you for your response. I Download http://diddywahdiddy.net/Puppy500/lupu-528.002_for_256MB.iso and make a frugal installation.
