How to password protect a single file or folder?

Is there a way to put a password on a single file or folder so every time someone tries to open it, they must enter a password?

If you don't encrypt the file or folder, then for true security you must prevent anyone from booting a live CD or USB flash memory stick in the computer, and accessing the protected file or folder that way. So I guess the obvious answer is, encrypt the file or folder you want to protect. I believe there is an encryption program for Puppy (truecrypt?) which allows you to encrypt single files or folders. To decrypt an encrypted file or folder, you enter a password and the program does the rest. That should be pretty close to what you want.
